Image Image Image Image Image Image
Fig. 1 Schematically represented A, B, and C fragments of recombinant Der p 2 comprising 126 amino acids for epitope analysis.
Fig. 2 SDS-PAGE analysis showing GST-fusion proteins of three dissected fragments (A, B, and C) of recombinant Der p 2 purified by using a GST-affinity column. Lane G represents GST.
Fig. 3 The reactivity of monoclonal antibodies against three dissected fragments and full-length recombinant Der p 2 was examined by ELISA. Monoclonal antibodies of 3B12, 2B6, and 4A5 as well as immune mouse serum showed reactivity to both B polypeptide and full-length recombinant Der p 2. However, a monoclonal antibody of 5D10 only reacted to full-length recombinant Der p 2.
Fig. 4 Immunoblot analysis showed that a monoclonal antibody (3B12) to recombinant Der p 2 reacted with only B-polypeptide (amino acid residues 44-93) of three dissected fragments of recombinant Der p 2.
Fig. 5 A control curve was produced for recombinant Der p 2 by two-site capture ELISA with monoclonal antibodies. The limit of sensitivity was 4 ng/ml.
Fig. 6 Control curves were produced for Dermatophagoides pteronyssinus and D. farinae crude extracts by ELISA. The limit of sensitivity of this assay was 8 µg/ml of D. pteronyssinus crude extract.
